  • In silico and in vitro analyses of a novel FoxO1 agonist reducing Aβ levels via downregulation of BACE1

    A potential of 3HFWC, a hydroxylated fullerenol derivative, to affect amyloid-b aggregation was evaluated in the 5xFAD transgenic mouse model of Alzheimer's disease (AD). The effect of the treatment was confirmed through NIRS analyses and applied machine learning. Our study showed that the presymptomatic, 3-month-long 3HFWC treatment significantly reduced the plaque load in the cortex of treated animals. At the same time, the treatment did not induce astrogliosis, microglial activation, and synaptic vilnerability. These data suggest that 3HFWC can be considered an adjuvant in the development of new therapeutic strategies in AD treatment.
